VIP — Vasoactive Intestinal Peptide

VIP is a 28-amino-acid neuropeptide of the secretin/glucagon superfamily, first isolated from porcine intestine. It is widely distributed across the central nervous system, autonomic ganglia, gastrointestinal tract, and immune tissues, where it functions as a neurotransmitter, neuromodulator, and immune regulator. VIP is among the most pleiotropic peptides in the research literature.

Mechanism of Action

VIP binds two G protein-coupled receptors, VPAC1 and VPAC2, activating Gs-coupled cAMP/PKA signaling and downstream CREB-mediated transcription. Research has characterized VIP’s role in smooth muscle relaxation via nitric oxide and cAMP mediators, bronchodilation, regulation of circadian rhythm in the suprachiasmatic nucleus, and shifting of immune responses toward Th2 and regulatory T-cell phenotypes. VIP also acts as a potent endogenous modulator of mitochondrial biogenesis and neuroprotection in CNS injury models.
